  1. Prepare the small parfait cup by thoroughly rinsing it with water to ensure cleanliness.

  2. Open the tissue culture container and carefully remove the plants, taking care not to damage them.

  3. Rinse all of the gel off the tissue culture plants thoroughly under running water. This step is crucial to remove any residual gel and ensure the plants are clean and free from contamination.

  4. Place the tissue culture plants into the parfait cup, ensuring they are not overcrowded and have space to breathe.

  5. Add three to four tablespoons of water into the parfait cup. This provides hydration to the tissue culture plants during the acclimation process.

  6. Add 2 ounces (approximately 60 milliliters) of Fluval Stratum into the parfait cup. Fluval Stratum is a substrate specifically designed for planted aquariums, providing essential nutrients for plant growth.

  7. Add a layer of sphagnum moss on top of the Fluval Stratum. Sphagnum moss helps retain moisture and provides a humid environment, which is beneficial for tissue culture plants during acclimation.

  8. Take a swab of Betadine and dilute it in 3 cups of water. Let the tissue culture plants soak in this Betadine solution for a few minutes before placing them in the substrate. Betadine is an antiseptic solution that helps prevent bacterial and fungal infections, ensuring the health of the plants during acclimation.

  9. Place the parfait cup in an appropriate environment for tissue culture plants. This environment should have sufficient light, preferably indirect or low light to prevent stress on the plants during acclimation. Additionally, maintain a stable temperature and humidity level to promote healthy growth.

  10. Monitor the tissue culture plants closely during the acclimation process. If you tug on the tissue culture plant gently and it doesn't come loose, it indicates that the plant is well-acclimated.

  11. After a period of acclimation, typically lasting 1-2 weeks, carefully transfer the tissue culture plants into their permanent aquarium or terrarium setup. Take care to acclimate them slowly to the new environment to minimize stress and ensure successful establishment.

By following these updated directions, you can effectively acclimate tissue culture plants using the Acclamation Kit, promoting their health and ensuring successful integration into their new environment.

Here's the additional step you can include after successful acclimation:

  1. After successful acclimation, you can consider changing the substrate to a mixture of soil, moss, coir, and orchard mix to provide further nutrients and support to the tissue culture plants. Aim for a chunkier mixture, as this will promote a healthier rooting system for the plants.

  2. Combine soil, moss, coir, and orchard mix in a ratio suitable for your specific plants' needs. Ensure that the mixture is well-draining yet retains moisture effectively.

  3. Carefully remove the tissue culture plants from the parfait cup, taking care not to damage their roots.

  4. Place the tissue culture plants into the new substrate mixture, ensuring that the roots are adequately covered and supported.

  5. Some enthusiasts choose to add clay balls to the substrate mix to enhance aeration and allow better penetration of air to the roots.

  6. Monitor the plants closely after the substrate change to ensure they adapt well to the new environment. Adjust watering frequency and light exposure as necessary to promote healthy growth.

By following these additional steps, you can further support the growth and development of tissue culture plants after successful acclimation, providing them with a nutrient-rich substrate and optimal conditions for thriving.
